Peter Parker is, in fact, the one and only son of Richard and Mary Parker. Captain Richard Parker and Mary Fitzpatrick had been agents in the organization that many of us know as S.H.I.E.L.D.
They were secret agents who went on a number of missions for the organization (in one comic-plot they were also the agents that freed a little known fella by the name of Logan a.k.a. Wolverine) - and were assassinated on the job.
Richard Parker had a brother named Ben, and Ben married May Reilly Parker - the raising guardians of your friendly neighborhood buddy.
